I would like to print some weatherboard walls to make some houses for a town scene.
I have found a HO scale house online (thingaverse) that will work perfectly (I'll modify the prints to eliminate uniformity).
My questions are; how do I scale up a HO model into a large scale one?
I have tinkercad just do not know how to use it correctly; is it just a matter of importing the stl files and changing the scale than saving the up scaled files to print?

Yes. STL files can be imported into Tinkercad provided they aren't too complex. They can then be resized by the requisite amount and then exported as new STL files.
Assuming the parts are the walls of the building which have been drawn flat, then you probably won't need to stretch them as much on the Z axis otherwise they will become too thick and chunky.

I have tried to rescale things downloaded from the internet a few times. It hasn't worked very well for me.
The problem is that if the model has been designed in a smaller scale the level of detail and accuracy may be limited - not visible in its designed scale - but very obvious when enlarged by a factor of 4. Might not be an issue but something too bear in mind.
Particularly likely to be an issue upscaling from 4mm scale to 16mms scale.

Trevor has answered above 4mm scale to 16mm scale is 400% or x 4.
HO to 16mm is x 5.4375
HO to 16mm is x 5.4375
